Dear all, I am looking to recruit two post-doctoral researchers experienced in cryo-EM, cryo-ET, or X-ray crystallography. The projects are focused on characterizing the replication of viruses in situ: We study the infection of enteroviruses by a combination of FIBM and cryo-ET to determine the mechanism of enterovirus genome delivery into a host cell and changes in intra-cellular organization induced by infection. We complement the whole-cell studies by high-resolution cryo-EM of virus genome release under biologically relevant conditions in vitro. We employ cryo-ET and light microscopy to characterize the mechanisms that tailed phages, potential phage therapy agents, employ to invade a biofilm and infect bacteria. We use the cryo-ET to determine the functions of bacterial factors in phage genome delivery and characterize phage assembly. We employ time-lapse fluorescence microscopy to establish the relationship between the stage of biofilm development and its susceptibility to phage infection.
